简介:Prof. Zhiwei Huang is Director of the Optical Bioimaging Laboratory in the Department of Biomedical Engineering, College of Design and Engineering, National University of Singapore (NUS). He is an internationally recognized leader in biomedical optics, biophotonics, and advanced optical imaging. His research focuses on developing label-free super-resolution and nonlinear optical microscopy, including coherent Raman scattering and multiphoton microscopy, for biomolecular imaging, as well as fiberoptic Raman spectroscopy and endoscopic imaging for early detection, diagnosis, and image-guided treatment of epithelial precancer and cancer. Prof. Huang pioneered Raman endoscopy and label-free super-resolution bioimaging technologies. He has published over 150 peer-reviewed papers in leading journals, including Nature Photonics, Light: Science & Applications, and Gastroenterology, and has delivered more than 100 plenary, keynote, and invited lectures worldwide. He has filed over 20 U.S. patents, with 10 licensed for commercialization. His IMDX technology was ranked among the Top 10 Medical Devices at MEDICA (Germany). He is an SPIE Fellow in recognition of his outstanding contributions to biomedical photonics and optical imaging.
讲座内容简介:High-speed stimulated Raman scattering (SRS) microscopy promises label-free visualization of cellular chemistry but remains constrained by shot-noise-limited sensitivity. In this seminar, I will present PHYSIQ-SRS, a physics-informed imaging framework that fundamentally redefines SRS acquisition using paired in-phase/quadrature measurements. By generating co-registered images with independent shot noise, PHYSIQ-SRS enables fully self-supervised denoising without ground truth or temporal redundancy, delivering ~12.5 dB SNR improvement while preserving quantitative chemical information. This advance enables video-rate volumetric Raman imaging and robust 4D (3D + time) tracking of lipid droplets in living cells. Using this capability, we uncover previously inaccessible metabolic phenotypes, revealing how intracellular transport is dynamically reprogrammed by nutrient availability, glycolytic inhibition, and cell division. PHYSIQ-SRS establishes a general solution for shot-noise-limited microscopy and paves the way for high-speed, label-free functional imaging across cell biology, metabolism, and biomedicine.
